Multiple returns is a waste of time. The return flow from your sump will be negligible compared to the output from a few appropriately sized powerheads -- especially once it's been divided in half and run through additional elbow fittings. It would be like blowing through a straw to try to change the direction of air from a 14" fan.
I'd just go with a center overflow and have the return come through the overflow

That's what I have, but I only run a single durso. Any concerns with a fourth hole in that location?

Umm... Well, I'm of the opinion that a Beanimal overflow is like putting a piece of duct tape over top of a screw to hold two pieces of wood together tighter. So there's that.
But no, I'd have no concerns regarding structural integrity if the holes are properly spaced out and the glass is of the appropriate thickness.
